“A two-component system controls the expression of genes encoding hemicellulases in Clostridium cellulolyticum”

Résumé :

The composition of the cellulosomes - a multi-enzymatic complex involved in the degradation of plant cell wall polysaccharides - produced by Clostridium cellulolyticum differs according to the growth substrate used by the bacterium. In particular, the expression of a cluster of 14 hemicellulase-encoding genes (called xyl-doc) seems to be induced by the presence of straw and not of cellulose. Interestingly, genes encoding a two-component regulation system were found upstream of xyl-doc and products of these genes were predicted to be involved in the regulation of the expression of xyl-doc genes by straw. First evidence for the involvement of the regulator in the expression of xyl-doc genes was illustrated by the analysis of cellulosome composition produced by the regulator overproducing strain when grown on cellulose. Ion-trap LC MS/MS, allowed the detection of the products of all xyl-doc genes and of the product of the gene at locus Ccel_1656 predicted to bear a carbohydrate binding domain targeting hemicellulose. RT-PCR experiments further demonstrated that the regulation occurs at a transcriptional level and that the transcription of all xyl-doc genes is linked. Analysis of the transcription of xyl-doc genes and of the gene at locus Ccel_1656 in a regulator knock-out strain and in this strain complemented by the regulator confirmed the involvement of the regulator in the expression of xyl-doc genes and of the gene at locus Ccel_1656 in response to straw. Electrophoretic Mobility Shift Assays with the purified regulator further demonstrated that the regulator binds to very conserved DNA regions located upstream of the first gene of xyl-doc gene cluster and upstream of gene at locus Ccel_1656.
